About AlphaLISA Technology:


AlphaLISA (Amplified Luminescent Proximity Homogeneous Assay) is a bead-based, no-wash homogeneous assay platform, enabling the detection of analytes and a dynamic range of biological interactions, including protein-protein, protein-peptide, and protein-small molecule interactions. This technology utilizes streptavidin-coated Alpha Donor beads and AlphaLISA Acceptor beads. Upon excitation, a singlet oxygen molecule is generated in the Donor bead that triggers a series of chemical reactions in the Acceptor bead, resulting in the emission of light at 615 nm, which is directly proportional to the amount of analyte or binding events within the sample. Alpha technology allows for the direct detection of molecules of interest in a homogeneous format, eliminating the need for time-consuming wash steps.



About AlphaLISA SureFire Ultra Technology:


AlphaLISA SureFire Ultra (ALSU) is a subset of AlphaLISA technology offered by Revvity. Similarly to AlphaLISA, Donor and Acceptor beads are used with this platform. In ALSU systems, Donor beads are coated with streptavidin to capture a biotinylated detection antibody, while Acceptor beads are coated with a proprietary CaptSure™ agent to immobilize the other detection antibody labeled with a CaptSure tag. In the presence of a target protein, the antibodies bring the Donor and Acceptor beads into proximity. Upon excitation at 680 nm, a photosensitizer within the Donor bead converts ambient oxygen into an excited singlet state, which diffuses up to 200 nm and produces a chemiluminescent reaction in the Acceptor bead, emitting light at 615 nm. If an Acceptor bead is not in proximity to a Donor bead, little to no signal is produced over the background.


About HTRF Technology:

 
HTRF (Homogeneous Time-Resolved Fluorescence) is a homogeneous, no-wash immunoassay platform used to detect and measure proteins and molecular interactions. The technology employs two antibodies labeled with distinct fluorophores that bind simultaneously to the same target antigen. Upon excitation of the donor fluorophore by a light source such as a laser or flash lamp, energy is transferred to the acceptor fluorophore via Fluorescence Resonance Energy Transfer (FRET). The acceptor then emits fluorescence at a specific wavelength, typically 665 nm. The magnitude of the FRET signal is proportional to the concentration of antigen-antibody complexes present in the sample, allowing for quantitative analysis without the need for wash steps.

- HTRF: Excels in studying molecular interactions, including protein-protein interactions, kinase activity, receptor-ligand binding, and epigenetic modifications, as well as biomarker and cytokine measurement.
